== Pharmacology == The drug potentiates the tropomyosin receptor kinases TrkA, TrkB, and TrkC with EC50Tooltip half-maximal effective concentration values of 382 nM, 295 nM, and ~330 nM, respectively. As a positive allosteric modulator of TrkA and TrkB, ACD856 potentiates the effects of brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF). In addition to the tropomyosin receptor kinases, ACD856 is also a similarly potent positive allosteric modulator of certain other receptor tyrosine kinases, including the insulin-like growth factor 1 receptor (IGF1R) and the fibroblast growth factor receptor 1 (FGFR1). However, its efficacies at the IGF1R and FGFR1 were much lower than at the TrkA, TrkB, and TrkC. In animals, ACD856 has been found to reverse scopolamine- and dizocilpine (MK-801)-induced memory impairment, to improve age-related memory deficits, and to have sustained antidepressant-like activity. It has also been reported to possess neuroprotective properties. In humans, the drug has been shown to cross the blood–brain barrier and to induce dose-dependent changes in electroencephalogram parameters. === Harm assessment === Professor David Nutt, former chair of the Advisory Council on the Misuse of Drugs (ACMD) in the UK has said in 2009, "people are better off taking ecstasy or amphetamines than those [drugs] we know nothing about" and "Who knows what's in [mephedrone] when you buy it? We don't have a testing system. It could be very dangerous[;] we just don't know. These chemicals have never been put into animals, let alone humans." Les King, a former member of the ACMD, has stated mephedrone appears to be less potent than amphetamine and ecstasy, but that any benefit associated with this could be negated by users taking larger amounts. He also told the BBC, "all we can say is [mephedrone] is probably as harmful as ecstasy and amphetamines and wait until we have some better scientific evidence to support that."

== Adverse reactions == Sleeping pills, including zopiclone, have been associated with an increased risk of death. The British National Formulary states adverse reactions as follows: "taste disturbance (some report a metallic taste); less commonly nausea, vomiting, dizziness, drowsiness, dry mouth, headache; rarely amnesia, confusion, depression, hallucinations, nightmares; very rarely light-headedness, incoordination, paradoxical effects [...] and sleep-walking also reported". Long-term use of Z-drugs, including zopiclone, has been associated with dependence, withdrawal symptoms, and cognitive impairment.

==== Diabetes ==== A meta-analysis reported that vitamin D supplementation significantly reduced the risk of type 2 diabetes for non-obese people with prediabetes. Another meta-analysis reported that vitamin D supplementation significantly improved glycemic control [homeostatic model assessment-insulin resistance (HOMA-IR)], hemoglobin A1C (HbA1C), and fasting blood glucose (FBG) in individuals with type 2 diabetes. In prospective studies, high versus low levels of vitamin D were respectively associated with a significant decrease in risk of type 2 diabetes, combined type 2 diabetes and prediabetes, and prediabetes. A systematic review included one clinical trial that showed vitamin D supplementation together with insulin maintained levels of fasting C-peptide after 12 months better than insulin alone.

In 2023, the US State Department announced plans to launch a "global coalition to address synthetic drug threats", with more than 80 countries expected to join. That April, Anne Milgram, head of the DEA since 2021, stated to Congress that two Mexican drug cartels posed "the greatest criminal threat the United States has ever faced." Pentoxyverine (rINN) or carbetapentane is an antitussive (cough suppressant) commonly used for cough associated with illnesses like common cold. It is sold over-the-counter as Solotuss, or in combination with other medications, especially decongestants. One such product is Certuss, a combination of guaifenesin and pentoxyverine. The drug has been available in the form of drops, suspensions and suppositories. It was formerly available over-the-counter in United States.
